*The best time to take sermorelin is prior to bedtime at least 2 hours after your last meal. WHAT IS SERMORELIN?
Sermorelin is a peptide that stimulates the release of human growth hormone releasing hormone (GHRH) from the hypthothalamus, which promotes the natural production and release of growth hormone (HGH) from the pituitary gland.
HGH is normally produced during our youth and helps build muscle, build bone, breakdown fat, and keep our skin young and healthy. As we get older, GH levels decline to the point that most people over age 50 have levels way below optimum. Directly administering GH can be tricky in that too much can be administered and does not induce the natural physiologic "pulse".
Sermorelin stimulates your own natural production of HGH, which works with our body's natural feedback mechanisms to prevent you from getting too much HGH. It's a cheaper and safer way to naturally restore your HGH levels.
SERMORELIN BENEFITS
While patients will notice some significant increases in changes in the body after the first month, the full benefits are usually fully noticed after 3-6 months of therapy. How and where in your body is HGH made?
Your pituitary gland is a small, pea-sized endocrine gland located at the base of your brain. It’s made of two lobes, the anterior lobe, and the posterior lobe. Your anterior pituitary lobe makes HGH. HGH is not released continuously throughout the day; instead, it is released in a number of “bursts” or “pulses” during the day. Peak HGH secretion occurs within an hour after the onset of deep sleep. This is one reason why getting a good night’s sleep is so important.
What triggers your pituitary gland to release HGH?
Your hypothalamus is the part of the brain that regulates many bodily functions. It is connected to your pituitary gland through a stalk of blood vessels and nerves (the pituitary stalk). Through this stalk, your hypothalamus communicates with your pituitary gland and tells it to release certain hormones.
In this case, your hypothalamus releases two hormones that affect HGH production. The first hormone is growth hormone-releasing hormone (GHRH). This hormone travels down the pituitary stalk from the hypothalamus to the anterior pituitary and tells it to release HGH by binding to special receptors. On the other hand, your hypothalamus also produces somatostatin, which travels down to the anterior pituitary and inhibits HGH from being released.
So why not just take HGH?
Scientists have developed a synthetic version of HGH. HGH therapy involves directly injecting this synthetic version into your body. However, this version behaves differently than your natural HGH. Your own HGH is much safer and more effective. It is also difficult to time injections to mimic the way your natural HGH is released in pulses. HGH therapy is very expensive and can even shut down your body’s natural production.
What is a better option?

Because sermorelin is closely related to naturally occurring GHRH, it can also stimulate the pituitary gland to release GH by binding to the same receptors as GHRH. This can have some excellent effects on the body such as increased lean body mass, fat reduction, improved energy, increased strength and endurance, accelerated wound healing, better sleep quality, etc.

What about Tesamorelin?
Tesamorelin is also a synthetic version of GHRH. Instead of using the first 29 amino acids of natural occurring GHRH as scientists did with Sermorelin, the developers of this peptide used all 44 amino acids and added a special attachment to the end to protect it from being degraded. Tesamorelin, like sermorelin, also binds to the natural GHRH receptors on the anterior pituitary gland and stimulates your body’s natural HGH release.
Tesamorelin was approved by the FDA in 2010 as a weight loss and fat reducing medication for HIV patients suffering from a condition called “lipodystrophy”, a condition that can cause an abnormal accumulation of fat in the abdominal area. Although it is not FDA-approved to treat non-HIV patients for this, research indicated that it would also benefit any patient suffering from excess abdominal body fat. It has recently gained popularity in this area.
Tesamorelin is considered to be the most effective hormone on the market to stimulate the natural production of HGH. It is also thought to be more effective in targeting belly fat. However, it is more expensive than sermorelin and is not very commonly compounded.

What about CJC-1295?
CJC-1295 is another similar medication. Like Sermorelin, it is made up of the first 29 amino acids of naturally occurring GHRH. However, some of the amino acids have been modified to extend the half-life compared to sermorelin, which is missing these modifications. They both act on the same GHRH receptors on the anterior pituitary and tell it to release HGH, but sermorelin mimics your body’s natural HGH pulses more closely because it’s cleared from the body faster than CJC-1295. You do not have to inject CJC-1295 as often though, since it has a longer half-life. It is often combined with ipamorelin for an added effect on natural HGH production.

What other ways can we naturally stimulate HGH secretion from the anterior pituitary? With ghrelin mimickers.
Ipamorelin is a synthetic five-amino acid chain growth hormone-releasing peptide (GHRP). It stimulates the natural HGH release from the anterior pituitary differently than the above medications. Instead of acting similarly to GHRH, ipamorelin stimulates the release of HGH by mimicking the actions of a gastric hormone called “ghrelin” in your body. Ghrelin is a hormone naturally found in your gastrointestinal system that binds to receptors on the pituitary gland.
This binding triggers the release of HGH. Ipamorelin increases your natural HGH release by mimicking ghrelin and binding to these ghrelin receptors. Ipamorelin also suppresses somatostatin. Somatostatin, if you recall, suppresses the release of HGH. So, we are suppressing the suppressor, if you will. Ipamorelin is the favored peptide of the GHRPs.
It is often paired with CJC-1295 for an additive effect of increasing natural HGH-production in more ways than one.

What about other ghrelin-mimickers?
Growth Hormone-Releasing Peptide 2 (GHRP-2) is another synthetic peptide that increases the release of growth hormone in the body and suppresses somatostatin production by mimicking ghrelin. We mentioned above that ipamorelin also does this. However, GHRP-2 is what’s referred to as a “non-selective growth hormone secretagogue” because it stimulates the release of HGH as well as other hormones in the body. Ipamorelin, however, only stimulates the release of GH, because it is a “selective secretagogue”.
Because of the less-selective nature of GHRP-2 on hormone release, it can have more side effects such as increased appetite and water retention. Although the two have similar efficacy in stimulating HGH-release in the body, many people prefer ipamorelin due to possibly less side effects. However, with ipamorelin being unavailable, it’s the “next-best” option.
GHRP-6 is a similar hormone that is older and considered less-potent than GHRP-2. It can stimulate the hunger response more than GHRP-2, which can benefit individuals who need to eat more (those who engage in intense physical training and endurance athletics). This makes it less desirable if you are trying to lose weight, however.
Both GHRP-2 and GHRP-2 are currently unavailable in the injectable form due to recent FDA restrictions passed in September 2023. This also applies to other ghrelin mimickers like ibutamoren (MK-677).
However, GHRP-2 is available in an oral form from both WP Pharma and Wells Pharmacy. It is called “Welltropin”.
So, what is best for me?
Before they were banned restricted by the FDA, a combination of CJC-1295 with Ipamorelin was one of the most popular combinations for naturally increasing HGH production. By mimicking both GHRH (CJC-1295) and ghrelin (ipamorelin), they worked together to enhance metabolism, aid in muscle growth and recovery, improve sleep, improve the appearance of your skin, increase energy, and many other benefits. However, with the new restrictions, we have to combine other medications from these two classes. Only sermorelin, tesamorelin, and Welltropin (GHRP-2) are available.
Combining either sermorelin or tesamorelin with Welltropin would be a great combination therapy. Tesamorelin is considered superior to sermorelin and thought to target belly fat more, but it’s more expensive, only available in an injection, and is only available from one pharmacy. However, a combo of sermorelin and Welltropin would still make an amazing addition to your weight loss journey. Plus, sermorelin is available in two oral forms for those who do not want to take injections 5 nights a week: a troche and a capsule combined with glutathione.
